Koleosho's football journey began at age 7 with Trumbull United in Connecticut. He then played for Manhattan Kickers from ages 7 to 11, where trips to Spain sparked his interest in developing there. In 2016, Koleosho joined the youth academy of FC Reus in Spain before moving to Espanyol's youth system in 2020. He progressed through Espanyol's youth ranks, signing his first professional contract on June 2, 2021. Koleosho's talent was evident as he helped Espanyol's U19 team reach the Copa del Rey Juvenil final in 2022. He also gained experience with Espanyol B in the fourth-tier Segunda División RFEF during the 2021-22 season.

Luca Koleosho's senior career began at Espanyol in Spain, where he made his professional debut on May 22, 2022, at the age of 17. Coming on as a substitute in the last match of the 2021/22 La Liga season against Granada, this appearance marked the beginning of his journey in top-flight football. In the 2022/23 season, Koleosho was promoted to Espanyol's first team. However, his progress was somewhat hindered by injuries.
